How to use sterile syringe filter?
- Before drawingthe sample, draw about 1ml of air into the syringe to minimize liquid retention.
- Draw the sample into the syringe, then draw 1ml of air, invert the syringe and wipe the tip clean of any residue.
- Use the Luer taperto connect the syringe to the needle filter, tighten gently to ensure a secure seal.
- Filter the solution in the syringe and inject it into the sample bottle, remove the filter, suck air into the syringe, then install the filter and press the pusher to filter out the residual sample, which can improve the sample recovery rate.

How to choose a suitable filter syringe?
Diameter
The most used sizes are 13mm and 25mm.
Choose the diameter according to the volume of sample:
For samples of 2~10mL, use 13mm;
For 10mL~100mL, use 25mm;
For samples larger than 100mL, consider 33mm.If the filtration volume is large, you can consider using a vacuum filtration bottle with a filter membrane for processing.
Pore size
0.2 um syringe filter can remove very fine particles in the sample and mobile phase as well as microorganisms. Can meet the requirements of GMP or Pharmacopoeia for sterilization 99.99%.
0.45 um syringe filter is for usually sample mobile phase filtration, capable of meeting general chromatographic requirements. For some environmental samples, biofuels and liquid samples with high particle content such as food and beverages, 0.45um filters can be used in conjunction with glass fiber filter paper.
For turbid solutions that are difficult to handle, they can be pre-filtered with a 1-5um membrane and then re-filtered with the appropriate membrane.
0.22 um syringe filter and 0.45 micron syringe filter are most used.
Material
PTFE syringe filter: Filtration of all organic solutions and strong acids and bases, especially strong solvents that are not tolerated by other membranes.
PVDF syringe filter: Hydrophobic PVDF membrane is mainly used in gas and vapor filtration, high temperature liquid filtration. Hydrophilic PVDF membrane is mainly used in the aseptic treatment of tissue culture medium and solution, and filtration of high temperature liquid.
Nylon syringe filter: Filtration of aqueous solutions and organic mobile phases.
PES syringe filter(polyethersulfone syringe filter): Designed for biochemical, laboratory, pharmaceutical and sterilization filtration.
MCE: Filtration of aqueous solutions or sterilization of heat-sensitive preparations.
Hydrophilic samples: It is appropriate to use hydrophilic membrane, suitable for filtering water-based solutions. Such as tap water, surface water, available filter membrane for water membrane (MCE), nylon membrane (Nylon).
Organic and inorganic compounds (alcohols, esters, oils) samples: suitable for nylon membrane.
Protein solution samples: low protein adsorption filter membrane, i.e. PVDF membrane.
Strong organic solvent samples: hydrophobic PTFE is applicable.
Ion chromatography: PES membranes are generally considered to be more suitable for the filtration of solutions with low inorganic ions.We also have GF, CA, CN, PP membranes for the filter.
What are the caveats of using a syringe filter sterile?
- Notice that the bag is sealed and kept, and make sure that the gloves and tools are clean when you take them, cross contamination is the most difficult thing to solve and deal with.
- When using it, you can’t use too much force, so as not to break the filter membrane and lose the original filtration effect. There are some matrix more complex sample solution, especially fat or large molecules of protein is not removed, or emulsified solution, in the process of over the membrane will be particularly difficult to plug the membrane can not be filtered, this time to change a new one and then filter it. Prevent emulsification or add some sodium chloride to remove emulsification, how to make the sample solution clarified without suspension is the goal of your research.
- Strictly select the appropriate membrane, the sample solution is organic phase, be sure to use organic filter membrane, aqueous phase of the selection of aqueous phase of the special membrane, the membrane box are marked, pay attention to the distinction between the 1:1 of the aqueous phase organic phase, the two membranes are filtered once, strictly control the entry of any impurities.
- After the sample solution with pigment passes through the filter membrane, the membrane becomes red, and the erythrosine is not found at all after feeding the sample into the machine, and then we know that the erythrosine is adsorbed by the filter membrane. Therefore, the needle filter must do experimental verification when filtering solvents with pigments, and choose the filter membrane that will not adsorb all pigments.
Can syringe filters be used for sterile filtration?
Yes. Please choose a syringe filter marked with “sterile” and operate in a sterile environment. It is often used in cell culture fluid filtration or microbiological experiments. When using it, you should also wear sterile gloves and use a sterile reagent bottle to collect the filtrate to avoid contamination.
