FAQ of Tissue Culture Tubes
How are sterile culture tubes used in the laboratory?
- Prepare liquid culture medium (sterile culture medium such as LB or M9 is recommended) and inject the required volume into the shake tube, not exceeding 50% of its total volume.
- Sterilization: Autoclave at 121℃ for 20 minutes, and make sure the lid is loose to avoid pressure accumulation.
- Inoculate strains: Use an inoculation loop to inoculate the strain into the shake tube under sterile conditions.
- Close the lid or use a breathable membrane: If aeration culture is required, a breathable sealing membrane can be used instead of an ordinary lid.
- Shaking culture: Place the shake tube in a constant temperature shaker and set the appropriate temperature and speed (such as 37℃, 180 rpm) for shaking culture.
- Subsequent operations: After the culture is completed, it can be transferred to a centrifuge tube for bacterial precipitation or further analysis.
How to use a cell culture tube during plasmid extraction?
After configuring the reagents and transformation, the next day, observe the colony growth. Then add medium to the culturing tube. Gently pick out individual colonies and place them in the shaker tube, then start the shaking process.
Can the round bottom culture tubes be used with other experimental equipment?
