- Speed and angle of shaking
The speed and angle of the shaking bottle is one of important factors that affect the results of the experiment.
If the shaking is too fast or too slow, it may affect the growth of the cells and the mixing of the culture solution.
The angle of the shaking needs to be adjusted based on the needs of the experiment to ensure that the culture solution can fully contact the cells when shaking.
2. Time and frequency
The time and frequency of shaking are also important points to note.
If shaking too short, the culture solution may not mix adequately, while shaking too long may cause overly vigorous cell movement, which may affect the growth and health of the cells.
The frequency of shaking also needs to be adjusted any time to ensure the culture solution can be mixed continuously.
3. Temperature and humidity control

What is the recommended shaker speed for plastic erlenmeyer flask?
Our recommended orbital shaker speed ranges from 75rpm-125rpm for different culture conditions.
The shaker speed required is generally determined by a number of factors such as the working volume of the shaker, the shaker amplitude, the type of cell line and medium, and whether the bottom of the shaker contains a baffle or not.
For example, if you use 5L erlenmeyer flasks with 25mm shaker amplitude to culture CHO cells or SF9 cells, we recommend a shaker speed of 90rpm.
What is the difference between a baffle bottom and a flat bottom conical flask?
The main difference between the two is the baffle at the bottom. The baffle design can effectively increase the gas exchange efficiency, especially at lower rotational speeds, the gas exchange efficiency is higher in the baffled bottom shaker.
When the culture volume is large, such as using 5L conical flask, in order to avoid producing too much bubbles under the higher speed and affecting the cell viability, it is necessary to reduce the speed, and the lower speed may reduce the efficiency of gas exchange, then you need to choose the triangular shaker with baffle design.
Difference between erlenmeyer flask and conical flask?
The main difference is in terminology: “conical flask” is the broader term used for any flask with a conical shape, while “Erlenmeyer flask” specifically refers to the conical flask design developed by Emil Erlenmeyer in the 1860s.
How to choose a proper volume for the erlenmeyer flask use?
Normally a conical erlenmeyer flask will be in a shaking shaker environment when used for cell culture. Too much space may cause the liquid splashing to the bottle mouth when shaking, increasing the risk of contamination, and compress the space for gas inside the bottle, which is bad for gas exchange during culturing. Too little working space may make the bottle medium liquid depth is insufficient, reducing the three-dimensional growth space of bacteria and cells. For erlenmeyer conical flask under 3L, we recommend the best working volume no more than the 30%-40% of the flask maximum volume.
